ZHO-1, an intrinsic MBL from the environmental Gram-negative species Zhongshania aliphaticivorans

Kieffer, Nicolas ; Guzmán-Puche, Julia ; Poirel, Laurent ; Kang, Hyo Jung ; Jeon, Che Ok ; Nordmann, Patrice

In: Journal of Antimicrobial Chemotherapy, 2019, vol. 74, no. 6, p. 1568–1571

Objectives: Our aim was to characterize the putative MBL of the environmental strain Zhongshania aliphaticivorans isolated from a marine environment. Methods: The putative MBL was identified in silico using the NCBI database. The β-lactamase gene was cloned into different Escherichia coli backgrounds. Kinetic parameters were determined using the purified enzyme. Results: The enzyme named ZHO-1...

Wide spread of carbapenemase-producing bacterial isolates in a Nigerian environment

Terrier, Christophe Le ; Masseron, Amandine ; Uwaezuoke, Nkolika Stella ; P.Edwin, Chinagozi ; E.Ekuma, Agantem ; Olugbeminiyi, Folake ; Shettima, Shuwaram ; Ushie, Simon ; Poirel, Laurent ; Nordmann, Patrice

In: Journal of Global Antimicrobial Resistance, 2020, vol. 21, p. 321–323

Objectives: The presence of carbapenemase-producing bacterial isolates is found not only in hospital and community settings but also in the environment. Carbapenemase production may be related to acquired, usually plasmid-borne, β-lactamase genes or to chromosomal genes intrinsic to various species. The aim of this study was to evaluate the occurrence of such carbapenemase-producing...

A Universal culture medium for screening polymyxin-resistant gram-negative isolates

Nordmann, Patrice ; Jayol, Aurélie ; Poirel, Laurent

In: Journal of Clinical Microbiology, 2016, vol. 54, no. 5, p. 1395–1399

The colistin-containing SuperPolymyxin medium was developed for screening polymyxin-resistant Gram-negative bacteria. It was evaluated with 88 polymyxin- susceptible or polymyxin-resistant cultured Gram-negative isolates. Its sensitivity and specificity of detection were ca. 100%. The SuperPolymyxin medium is the first screening medium that is able to detect intrinsic and acquired...

The type I-E CRISPR-Cas system influences the acquisition of bla KPC-IncF plasmid in Klebsiella pneumonia

Zhou, Ying ; Tang, Yu ; Fu, Pan ; Tian, Dongxing ; Yu, Lianhua ; Huang, Yunkun ; Li, Gang ; Li, Meng ; Wang, Yong ; Yang, Zehua ; Xu, Xiaogang ; Yin, Zhe ; Zhou, Dongsheng ; Poirel, Laurent ; Jiang, Xiaofei

In: Emerging Microbes & Infections, 2020, vol. 9, no. 1, p. 1011–1022

Klebsiella pneumoniae carbapenemase (KPC)-producing K. pneumoniae (KPC-KP) have disseminated worldwide and emerged as major threats to public health. Of epidemiological significance, the international pandemic of KPC-KP is primarily associated with CG258 isolates and bla KPC-IncF plasmids. CRISPR-Cas system is an adaptive immune system that can hinder gene expansion driven by horizontal gene ...

Transposition of tn125 encoding the ndm-1 carbapenemase in Acinetobacter baumannii

Bontron, Séverine ; Nordmann, Patrice ; Poirel, Laurent

In: Antimicrobial Agents and Chemotherapy, 2016, vol. 60, no. 12, p. 7245–7251

The blaNDM-1 gene encodes a carbapenemase that confers resistance to almost all β-lactams, including last-resort carbapenems. This is increasingly reported worldwide in nosocomial and community-acquired Gram-negative bacteria. Acinetobacter baumannii is an important opportunistic pathogen that is considered an intermediate reservoir for the blaNDM-1 gene. In this species, the blaNDM-1 gene...

Transposition of Tn1213 encoding the PER-1 Extended Spectrum β-lactamase

Mancini, Stefano ; Poirel, Laurent ; Kieffer, Nicolas ; Nordmann, Patrice

In: Antimicrobial Agents and Chemotherapy, 2018, p. -

PER-1 is an extended-spectrum β-lactamase that is encoded by a gene located into the composite transposon Tn1213 made by two distinct insertion sequences, namely ISPa12 and ISPa13. In vitro mobilization performed in Escherichia coli shows that Tn1213 is functional and is able to mobilize the blaPER-1 gene although at a very low frequency (ca 1x10-9).

Structure of the catalytic domain of the colistin resistance enzyme MCR-1

Stojanoski, Vlatko ; Sankaran, Banumathi ; Prasad, B. V. Venkataram ; Poirel, Laurent ; Nordmann, Patrice ; Palzkill, Timothy

In: BMC Biology, 2016, vol. 14, p. 81

Due to the paucity of novel antibiotics, colistin has become a last resort antibiotic for treating multidrug resistant bacteria. Colistin acts by binding the lipid A component of lipopolysaccharides and subsequently disrupting the bacterial membrane. The recently identified plasmid-encoded MCR-1 enzyme is the first transmissible colistin resistance determinant and is a cause for concern for...
