Antimicrobial activity of octenidine against multidrug-resistant Gram-negative pathogens

Alvarez-Marin, R. ; Aires-de-Sousa, Marta ; Nordmann, Patrice ; Kieffer, Nicolas ; Poirel, Laurent

In: European Journal of Clinical Microbiology & Infectious Diseases, 2017, p. 1–5

Multidrug-resistant (MR) Gram-negative (GN) pathogens pose a major and growing threat for healthcare systems, as therapy of infections is often limited due to the lack of available systemic antibiotics. Well-tolerated antiseptics, such as octenidine dihydrochloride (OCT), may be a very useful tool in infection control to reduce the dissemination of MRGN. This study aimed to investigate the...

Increased resistance to carbapenems in proteus mirabilis mediated by amplification of the bla vim-1 -carrying and is 26 -associated class 1 integron

Bontron, Séverine ; Poirel, Laurent ; Kieffer, Nicolas ; Savov, Encho ; Trifonova, Angelina ; Todorova, Iva ; Kueffer, Gwendoline ; Nordmann, Patrice

In: Microbial Drug Resistance, 2019, vol. 25, no. 5, p. 663–667

The aim of the study was to decipher the mechanisms and associated genetic determinants responsible for increased carbapenem resistance among Proteus mirabilis clinical isolates. Methods: The entire genetic structure surrounding the β- lactam resistance genes was characterized by PCR, gene walking, and DNA sequencing. Results: A series of clinical P. mirabilis isolates were consecutively ...

Colistin resistance in Parisian inpatient faecal Escherichia coli as the result of two distinct evolutionary pathways

Bourrel, Anne Sophie ; Poirel, Laurent ; Royer, Guilhem ; Darty, Mélanie ; Vuillemin, Xavier ; Kieffer, Nicolas ; Clermont, Olivier ; Denamur, Erick ; Nordmann, Patrice ; Decousser, Jean-Winoc ; The IAME Resistance Group

In: Journal of Antimicrobial Chemotherapy, 2019, vol. 74, no. 6, p. 1521–1530

Beyond plasmid-encoded resistance (mcr genes) prevalence in strain collections, large epidemiological studies to estimate the human burden of colistin-resistant Escherichia coli gut carriage are lacking. Objectives: To evaluate the prevalence of colistin-resistant E. coli carriage in inpatients and decipher the molecular support of resistance and the genetic background of the strains.Methods:...

In: Clinical Infectious Diseases, 2020, vol. 70, no. 9, p. 1998–2001

In July 2017, a patient presented colonization with a multidrug-resistant, carbapenemase (KPC-3)-producing Klebsiella pneumoniae isolate. A custom-made, lytic bacteriophage preparation was administered to the patient in December 2017, with subsequent eradication of the microorganism and without adverse effects.

The inoculum effect of Escherichia coli expressing mcr-1 or not on colistin activity in a murine model of peritonitis

Fantin, B. ; Poujade, J. ; Grégoire, N. ; Chau, F. ; Roujansky, A. ; Kieffer, Nicolas ; Berleur, M. ; Couet, W. ; Nordmann, Patrice

In: Clinical Microbiology and Infection, 2019, vol. 25, no. 12, p. 1563.e5-1563.e8

Colistin often remains the last resort antibiotic active against carbapenemase- producing Enterobacteriaceae. However, while in vitro inoculum effect has been reported, therapeutic relevance of this phenomenon remains questioned. Methods: Ten E. coli strains were used that included the wild-type CFT073 and its transconjugant CFT073-MCR-1 and eight susceptible clinical isolates. Mice with ...

Evaluation of the Rapid Polymyxin NP test and its industrial version for the detection of polymyxin-resistant Enterobacteriaceae

Jayol, Aurélie ; Kieffer, Nicolas ; Poirel, Laurent ; Guérind, François ; Güneser, Deniz ; Cattoir, Vincent ; Nordmann, Patrice

In: Diagnostic Microbiology and Infectious Disease, 2018, vol. 92, no. 2, p. 90–94

The commercial Rapid Polymyxin NP test was evaluated to detect colistin-resistant Enterobacteriaceae. A total of 223 enterobacterial isolates corresponding to 136 resistant (including 38 MCR-like producers), 19 heteroresistant, and 78 colistin- susceptible isolates were tested. The test was performed according to the manufacturer's instruction, and the color of the wells was read after 2 and 3...

High prevalence of carbapenemase-producing enterobacteriaceae among hospitalized children in luanda, angola

Kieffer, Nicolas ; Nordmann, Patrice ; Aires-de-Sousa, Marta ; Poirel, Laurent

In: Antimicrobial Agents and Chemotherapy, 2016, vol. 60, no. 10, p. 6189–6192

This study aimed to evaluate the prevalence of carbapenemase-producing Enterobacteriaceae in Luanda, Angola. A total of 157 rectal samples were collected from children visiting a pediatric hospital in Luanda in March 2015. Fifty-seven imipenem-nonsusceptible enterobacterial isolates were recovered, most of which were non-clonally related. The blaOXA-181 (50/57) and blaNDM-1 (7/57)...

Moraxella species as potential sources of MCR-like polymyxin-resistance determinants

Kieffer, Nicolas ; Nordmann, Patrice ; Poirel, and Laurent

In: Antimicrobial Agents and Chemotherapy, 2017, p. AAC.00129-17

Plasmid-mediated resistance to polymyxins mediated by the MCR-1/-2 determinants has been recently reported in Enterobacteriaceae worldwide. Using PCR-based and cloning strategies, a series of Moraxella species were screened for mcr-like genes. Moraxella spp. that are mainly animal pathogens but may also be human pathogens were identified as potential reservoirs of mcr-like genes.
